The Peppermint Bay Cruise and Hotel is fully wheelchair accessible. The vessel departs from Brooke St Pier, which is accessible via a ramp. The walkway on the vessel is reasonably narrow, so I am not sure how wider wheelchairs would go fitting through this area, however I had no trouble with my smaller size electric wheelchair.
